Peptide Tag Folding for Homodimer

Post by FabJu »

Hi there,
I created a recombinant variant of a homodimeric protein by attachment of 20 AA-peptide tag. Now I try to predict the structure of the dimer. The structure of the wildtype dimer is solved and the template is available.
Using LOMETS server I was able to model the structure of the monomer. But superimposing the modelled monomers with the dimer template leads to overlap of the attached peptide tags of both subunits?

Is there any chance to model the homodimer (incl. attached peptide sequence) directly based on the dimeric wildtype template?

Thanks,
FabJu

I think you can try to use TACOS https://zhanglab.dcmb.med.umich.edu/TACOS/
