Dear Zhang Lab,
I have a protein with quite low coverage of template, and I want to predict its 3D structe with good quality. It has a good coverage only at the domain side, and PDB proteins that cover the domain does not cover the whole protein. Therefore, the rest of it has no template to fold according to.
I was wondering if i-TASSER does TBM to this domain side and ab initio FM to the rest of the protein at the same time? If so, how can I do it? Also, how to do only ab initio FM at i-TASSER?

Dear user,
You are encouraged to use
https://zhanggroup.org/DMFold/
or
https://zhanggroup.org/D-I-TASSER/
servers.
When using D-I-TASSER, please try to select those three options.
Using large IMG/JGI metagenomic database to build MSA (additional one or two days maybe applied).
Using AlphaFold2 distances (additional one or two days maybe applied).
Using domain partition and assembly module (additional one or two days maybe applied)
Let us know if you have any questions.
Best Regards
Wei Zheng
